RESUMO

This work is a continuation of research of the Chizhevsky-Velhover's bio-astronomic effect. Monitoring of volutin granule metachromatic staining of Saccharomyces yeasts (S. cerevisiae UCM Y-517, S. cerevisiae CRY, S. cerevisiae CNX, S. bayanus UCM Y-493, S. unisporus UCM Y-2065, S. rosinii UCM Y-2614, S. exiguus UCM Y-649) under conditions of different space weather was carried out. S. cerevisiae UCM Y-517, which displayed the metachromatic reaction in 96.7 % cases, showed the biggest sensitivity to the space weather changes. The changes in phosphoric metabolism of S. cerevisiae CNX cells, which can not synthesize exopolyphosphatases PPX1 and PPN1 (CF 3.6.1.11), did not influence the metachromatic reaction. Yeast cells grown on wort-agar displayed more intensive metachromatic reaction compared to those grown on YEPD-agar. However, increasing concentration of phosphorus in YEPD-agar improved visualization of the metachromatic staining. The strain S. cerevisiae UCM Y-517 is recommended as a model for monitoring of volutin granules metachromatic reaction in the research project "Heliomed" because of its high sensitivity to the space weather changes.

In this work, the impact of space weather on the staining and the motility of volutin granules ("dancing bodies") in vacuoles of yeast Saccharoniyces cerevisiae with normal and changed phosphoric metabolism was studied. It was showed that the installed ear- lier relation between metachromatic reaction of volutin granules and galactic cosmic rays was confirmed by DAPI-staining of inorganic polyphosphates in cells S. cerevisiae UCM Y-517. The inverse correlation of efficiency of DAPI-staining of volutin granules with some solar activity indexes was observed. During the period of study, the rhythmicity of motile volutin granules in the studied cultures (S. cerevisiae UCM Y-517, S. cerevisiae CRY, S. cerevisiae CNX) was marked. The strains S. cerevisiae UCM Y-517 and CRY, both with unmodified phosphoric metabolism, showed the same rhythmicity of mobile volutin granules. However, this rhythmicity in the strain CNX, which cannot synthesize exopolyphosphatases PPX1 and PPN1 (CF 3.6.1.11), was changed. The volutin granule motion coincided with the rhythmicity of galactic cosmic rays within the period of 9 days. It can be suggested that "dancing bodies" are synchronized with rhythms of galactic cosmic rays. Observed differences between mutant and parental strains may indirectly indicate participation of polyphosphates in the reaction of cells on the changes of space weather pa- rameters. Thus, correlating variability of staining and motion of polyphosphate-containing volutin granules under action of cosmophysical factors may be an evidence of possible key role of phosphoric metabolism in sensitivity of yeast cells to space weather changes.

Inactivation of the key genes, which are responsible for the enzymes of polyphosphates degradation, exopolyphosphatases ppx1 and ppn1, caused both an increase of polyphosphates content in Saccharomyces cerevisiae cells and an increase in chain length of acid-soluble and alkali-soluble fractions. It had no effect on the frequency of volutine granules metachromasy that was based on the interaction of dye molecules with ionic groups of polyphosphates. At the same time, a mutant strain reaction to nystatin differed from the reaction of the parental and wild-type strains when the metachromasy was absent. Obtained data may indicate a pleiotropic effect of ppx1 and ppn1 genes, which encode the major exopolyphosphatase of Saccharomyces cerevisiae and affect the reaction of cells to external factors through changes in the metabolism of polyphosphates.

RESUMO

A possibility of purification of the surface storm drains from petroleum has been shown. The developed extensive biotechnology is based on the use of the preparation "Desna"--an active destructor of hydrocarbons. The application of the biotechnology at the plants for treatment of the surface storm waters from the industrial zone Telychka of the city of Kyiv has allowed the content of petroleum in water dropped to the Dnieper to be constantly reduced 50-100 times.
